Pressurized intraperitoneal aerosol chemotherapy with cisplatin and doxorubicin or oxaliplatin for peritoneal metastasis from pancreatic adenocarcino…

2020

Background: Systemic chemotherapy for pancreatic adenocarcinoma (PDAC) and cholangiocarcinoma (CC) with peritoneal metastases (PM) is affected by several pharmacological shortcomings and low clinical efficacy. Pressurized intraperitoneal aerosol chemotherapy (PIPAC) is expected to maximize exposure of peritoneal nodules to antiblastic agents. This study aims to evaluate safety and efficacy of PIPAC for PM of PDAC and CC origin. Methods: This is a retrospective analysis of consecutive PDAC and CC cases with PM treated with PIPAC at two European referral centers for peritoneal disease. We prospectively recorded from August 2016 to May 2019 demographic, clinical, surgical, and oncological data…

Polyaspartamide based hydrogel with cell recruitment properties for the local administration of hydrophobic anticancer drugs

2019

Abstract By exploiting the chemical versatility and the high water dispersibility of α,β-poly(N-2-hydroxyethyl)D,L-aspartamide, in this work, two different polymer derivatives were synthesized for the first time. Obtained macromolecules were characterized and used to produce hydrogels exploitable for the local release of hydrophobic anticancer drugs. The first derivative, bearing pendant β-cyclodextrins, was employed to solubilize tamoxifen, chosen as a model drug, and to produce a water soluble supramolecular complex, as evidenced through tamoxifen phase solubility studies. The second derivative, bearing pendant Cyclo(Arginine-Glyicine-Asparagine-D-Phenilyalanine-Cysteine) peptide moieties…

Pressurized Intraperitoneal Aerosol Chemotherapy (PIPAC) for Unresectable Peritoneal Metastases. Feasibility, Safety and Efficacy Outcomes

2022

Recently, a novel method for chemotherapy administration inside the abdominal cavity gained wide attention among surgical oncologists dealing with peritoneal surface malignancies. Based on laparoscopy, Pressurized Intraperitoneal Aerosol Chemotherapy (PIPAC) is a drug delivery system designed to overcome the known hurdles of intraperitoneal chemotherapy. Preclinical data suggest that the aerosol drug that PIPAC creates and the increased intraabdominal pressure obtained during laparoscopy provide highly effective distribution of cytotoxic compounds into tumor nodules. Several studies have documented the favorable safety profile and promising clinical outcomes of repetitive PIPAC in different…
